Experimental Hematology: Young Investigator articles of 2023


Experimental Hematology has reviewed articles published in 2023.  Among the articles highlighted are two Young Investigator articles that both explore important biological questions in normal and malignant hematopoiesis using murine models.

Rydström et al investigate how the hematopoietic stem cells regenerate after transplantation. The process where quiescent hematopoietic stem cells start to proliferate to replenish the hematopoietic system, and how they gradually return to homeostasis and quiescence is studied. To capture the dynamics of the process, HSPCs were transplanted and 8 different timepoints surveyed. The repopulating cells were studied at the transcriptional and functional levels. Differential expression of cell cycle genes and metabolic changes were observed during the process. The authors also investigate the functional capacity of the regenerating HSCs and found that it increases with time. Altogether, the study generates a detailed map of regeneration of HSPCs after transplantation.

The article by Flores et al, investigates how mutations in TET2 (Ten-eleven translocation) are affected by age in murine models. Somatic TET2 mutations are frequently associated with hematological malignancies. It has both catalytic and noncatalytic functions, and roles in myeloid and lymphoid lineages have been previously reported. The authors use two different loss of function models: a Tet2 catalytic mutant model and a Tet2 knockout model. Bone marrows from 3 to 12-month-old mice were transplanted and the recipient monitored for disease development. The Tet2 catalytic mutant cells caused mainly myeloid defects, irrespective of age. Tet2 knock out cells on the other hand, gave rise to both myeloid and lymphoid defects at early age, whereas at later time-points primarily myeloid defects were observed. This was in line with the molecular data, where a shift from lymphoid to myeloid dysregulation of genes could be seen with increased age of the transplanted cells. The study highlights developmental aspects of Tet2 catalytic and non-catalytic functions.
